Most bacteria in the species S. enterica belong to one of seven subspecies; all but subspecies I normally grow only in cold-blooded animals. Subspecies IIIb (S. Diarizonae) is naturally found in reptiles, and is capable of causing disease in humans but there are only about 10 reports per year in the USA. However, this serovar is often found in sheep in Europe.
Salmonella is a Gram-negative bacterium, a cylindrical rod of size about 2 microns by 0.5 microns, thus much smaller than the cells of higher organisms such as humans. In culture, the bacterium will grow on artificial media to form colonies. Subspecies IIIb (also called Diarizonae) is thought to be the first Salmonella to have originated from the flagellar monophasic strains such as S. Arizonae, and become biphasic for flagellar synthesis, i.e., it makes two types of flagella./p>
The strain of S. Diarizonae (61:1,v:1,5,(7)) being sequenced is CDC 01-0005. It was isolated from the stool of a 37 year old human female in Indiana, USA. It is maintained in the American Type Culture Collection as ATCC BAA-639 and in the Salmonella Genetic Stock Centre as SGSC4692. The genome is being sequenced to 8X coverage, using plasmid and fosmid libraries, and will be finished to an error rate of less than 1 per 10,000 bases. Automated annotation will be performed and manual annotation will continue in the labs of Michael McClelland and Kenneth Sanderson.
